Inhibition of Cancer Stem Cells Growth with Silibinin Encapsulated in Nanoparticles with Deregulation of miR-34a, miR-221, and miR-222

Background: Colorectal Cancer (CRC) is the most common malignant gastrointestinal cancer. stem cells (CSCs) are major cause of cancer recurrence and drug resistance. Silibinin, as an herbal compound, has anticancer properties. Objectives: The present study aimed to evaluate antiproliferative effects silibinin on HT29 stem-like (spheroids). Methods: In this study, apoptotic properties Silibinin encapsulated in Polymersome Nanoparticles (SPNs) were evaluated by MTT assay, propidium iodide (PI) /AnnexinV cell cycle analysis, DAPI (4',6-diamidino-2-phenylindole) staining. expression some miRNAs their potential targets was real-time reverse transcription-polymerase chain reaction (qRT-PCR). Results: IC50 SPNs determined at 28.13±0.78µg/ml after 24 h. (28µg/ml) induced apoptosis 32.36% staining indicated a decrease stained nuclei induction. treatment increased miR-34a, well P53, BAX, CASP9, CASP3, CASP8. downregulation miR-221 miR-222 observed treated cells. Moreover, level CD markers spheroids (cancer cells) compared untreated spheroids. Spheroids completely destroyed 72 h with (28µg/ml). Conclusion: As evidenced obtained results, can be used effective agent multi-layer mono-layer cancerous upregulation tumor suppressive miRs genes, oncomiRs oncogenes.
